Biogenesis of mitochondria relies on import of over 1000 different proteins from the cytosol. The majority of them are synthesized with N-terminal presequences and use the TOM and TIM23 complexes in the outer and inner mitochondrial membranes, respectively, to reach the final place of their function within the organelle. We use budding yeast as a model system and combine biochemistry, cell biology and genetics with biophysics and structural biology to elucidate the molecular mechanisms of protein translocation along the presequence pathway. Read more...
