We aim to elucidate the cellular interactions and molecular pathways that orchestrate the generation of a 'useful' T cell repertoire and imprint self-tolerance. To do so, we employ genetically manipulated mouse models that allow us to dissect T-cell-intrinsic and -extrinsic determinants shaping the T cell pool during repertoire selection in the thymus.
